The 2005 Chevrolet Corvette C6.R is a grand tourer racing car built by Pratt & Miller as well as General Motors for competition in endurance racing.
Most of the street legal Corvette (C6) design remains in the C6.R as they were designed alongside each other. Most of the retained design resulted in the increased use of lightweight aluminium and a better designed airflow across the body.
The C6.R saw an innovation in design with the use of an air conditioning system that was better suited to keeping drivers cool at high cockpit temperatures.
A development car was built on a C5-R chassis but adapted to carry C6.R bodywork. The GT1 version debuted in 2005 followed by multiple American Le Mans Series championships, Le Mans Series, FIA GT Championship and 24 Hour of Le Mans victories.

The performance of the C6.R is up to par with the fastest cars due to its quick acceleration and good handling.

The stock Corvette C6.R is unlocked upon the player achieving 100% game completion across both the Career mode and the Challenge Series.
Upon being unlocked, it is made available as a Bonus car for use in Quick Race or multiplayer. It can not be purchased in the Career mode or be selected for customisation in My Cars.

Challenge Series
A custom Corvette C6.R is offered in the Challenge Series and can be driven in two events:
- #57 - Tollbooth Time Trial: Complete "Beacon & Petersburg" in less than four minutes and thirty-five seconds.
- #58 - Roadblocks: Dodge ten police roadblocks in a Heat Level 3 pursuit and evade.
It can not be unlocked for use in any events or game modes outside of its two Challenge Series events.

- In Need for Speed: Most Wanted, Sergeant Cross' car differs between cutscenes and gameplay. His Corvette featured in cutscenes is a C6.R, although the car used during in-game pursuits is a variant of the Corvette Federal Cruiser.
